Devil Hunter to get used to the controls and combat. Human enemies are too fragile to even do that. Id recommend Son of Sparda or even Dante Must Die for the challenge, but you need to go out of your way to get that for your first playthrough and you'd probably be unhappy at the controls while doing DMD difficulty for the first time anyways. #2.
